测序级重组赖氨酰内肽酶/胰蛋白酶预混装

◆概述


本品选用A. Lyticus 来源氨基酸序列,利用独特大肠杆菌重组表达技术制备的Lys-C,能够特异性识别并酶切肽链中Lys羧基端肽键;其与重组表达并经甲基化修饰的测序级胰蛋白酶,经研究和方法验证,按照特定比例混合后,能够在一次酶切或分步酶切中,保证特异性的前提下,酶切效率更高,便于进行序列分析。

 

◆优势


1)采用甲基化修饰重组表达胰蛋白酶,其赖氨酸残基经甲基化修饰有效防止酶切缓冲中胰酶自切或被Lys-C降解造成胰酶活性的降低。

2)独有的Lys-C重组表达制备技术,保证高纯度与高酶活性的同时,无任何杂酶活性,确保酶切反应的特异性。

 

◆纯度·来源


选用A. Lyticus 来源氨基酸序列,利用独特大肠杆菌重组表达技术制备的Lys-C

选用猪的胰腺来源阳离子型胰蛋白酶基因,利用毕赤酵母菌株重组表达技术制备的胰蛋白酶

 

◆使用方法


溶解缓冲:用无菌水或者50 mmol/L Tris-HCl,pH 8.0~8.5;溶解后分装,-15℃以下保存。

酶切缓冲:50 mmol/L Tris-HCl,pH 8.0~8.5

推荐条件:蛋白酶:融合蛋白质量比1:25~1:100(蛋白组学或肽图)37℃反应。

          针对疏水性强、溶解性较差的蛋白,可添加变性剂或者封闭二硫键(烷基化)使其空间结构被打开,从而提高酶切效率或者提高肽图覆盖率。

可以参考以下方法:


两步溶出法消化:可以添加终浓度为5 mmol/L的DTT 在室温反应30 min ; 再加入终浓度为10 mmol/L 的碘乙酰胺,在室温避光反应30 min。蛋白溶液中补入终浓度6~8 mol/L 尿素,胰蛋白酶/Lys-C 与蛋白质或蛋白质混合物以1:25 的蛋白质∶蛋白酶比(w/w)混合并在37℃下孵育3~4 h,用酶切缓冲稀释尿酸降至1 mol/L 以下,在37℃反应过夜。

终止反应:加入终浓度0.5-1%三氟乙酸,14000-16000×g,离心10 min 去除沉淀颗粒。

 

◆保存条件


储存稳定性:冻干粉存于-15℃以下,24 个月稳定;50 mmol/L Tris-HCl pH 8.0~8.5 溶解后储存于-15℃以下,反复冻融5次,无活性损失。

运输稳定性:冰袋保温运输,活性稳定。

Tri-Methyl-Histone H3 (Lys9) (D4W1U) Rabbit mAb detects endogenous levels of histone H3 when tri-methylated on Lys9. This antibody shows some cross-reactivity with histone H3 that is di-methylated on Lys9, but does not cross-react with non-methylated or mono-methylated histone H3 Lys9. This antibody does not detect tri-methyl histone H3 Lys9 when the adjacent Ser10 residue is phosphorylated during mitosis. In addition, this antibody does not cross-react with methylated histone H3 Lys4, Lys27, Lys36, or Lys79.Monoclonal antibody is produced by immunizing animals with a synthetic peptide corresponding to residues near the amino terminus of histone H3 in which Lys9 is tri-methylated.Species predicted to react based on 100% sequence homology: Bovine.
