Amplite 荧光法肾素检测试剂盒 红色荧光 是美国AAT Bioquest生产的用于β-半乳糖苷酶的试剂盒，肾素是一种酶，参与人体的肾素-血管紧张素系统（RAS），介导细胞外体积和动脉血管收缩。它调节血压和电解质的稳态。血管紧张素II收缩血管，导致血压升高。它还会增加ADH和醛固酮的分泌，并刺激下丘脑激活口渴反射。肾素-血管紧张素系统过度活跃会导致血管收缩以及钠和水的滞留。肾素已被确定为治疗高血压的有吸引力的靶标。 Amplite 肾素检测试剂盒使用我们专有的Tide Fluor 3（TF3）/ Tide Quencher 3（TQ3）荧光共振能量转移（FRET）肽，为高通量筛选肾素抑制剂和肾素活性提供了便捷的检测方法。在FRET肽中，TF3的荧光被TQ3淬灭。通过肾素裂解为两个单独的片段后，TF3的荧光得以恢复，荧光信号可以通过荧光酶标仪轻松监测。该测定法比基于EDANS / DABCYL的测定法灵敏度高约五十倍。借助Amplite 肾素检测试剂盒，我们在100 µL反应体积中检测到低至1ng的肾素。但是，试剂盒中使用的肾素底物的选择性尚未经过彻底测试。由于基于肽的蛋白酶底物通常具有低选择性，因此它也可能对其他蛋白酶有反应。金畔生物是AAT Bioquest的中国代理商，为您提供最优质的Amplite 荧光法肾素检测试剂盒 红色荧光 。

产品说明书
样品实验方案
简要概述
1.添加适当的对照或测试样品（50 µL）
2.加入Renin Red 底物工作溶液（50 µL）
3.在37°C的培养箱中孵育30-60分钟（用于终点读数）
4.监测Ex / Em = 540/590 nm的荧光强度
溶液制备
1.储备溶液
肾素标准
向487.5 µL的测定缓冲液（组分C）中加入12.5 µL的40 µg / mL肾素标准溶液（组分B），得到1000 ng / mL的Renin标准溶液（Ren7）。 取150 µL 1000 ng / mL Renin标准溶液进行1：3连续稀释，以得到连续稀释的Renin标准液（Ren6- Ren1）。
2.工作溶液
将50μLRenin Red 底物（组分A）添加到5 mL的测定缓冲液（组分C）中，使总体积为5.05 mL。 注意：应立即使用Renin Red 底物。

1.根据需要准备含有肾素的生物样品。
2.根据表1和表2中提供的布局准备肾素标准品和/或含肾素的测试样品。对于384孔板，每孔使用25 µL试剂代替50 µL。
3.如果您正在筛选肾素抑制剂，则将板在酶反应所需的温度（例如25°C或37°C）下预孵育10-15分钟。
4.将50 µL（96孔）或25 µL（384孔）的Renin Red 底物工作溶液添加到样品和测定板的对照孔中。
5.将反应在37°C的培养箱中孵育30至60分钟。
6.使用荧光板读数器在Ex / Em = 540/590 nm（截止= 570 mn）下监控荧光强度。注意：试剂盒中使用的肾素底物的选择性尚未经过彻底测试。由于基于肽的蛋白酶底物通常具有低选择性，因此它也可能对其他蛋白酶有反应。人们可能使用肾素特异性抑制剂进行特异性测试，例如在存在肾素特异性抑制剂的情况下，底物的水解仅是由于非特异性蛋白酶活性。总活性和在肾素特异性抑制剂存在下的活性之间的差异给出了样品中的肾素活性。
对于动力学读数：立即开始测量荧光强度，并每5分钟连续记录数据30至60分钟。
对于终点读数：将反应液在37°C下孵育60分钟或更长时间，如果可能，请避免光照。 然后测量荧光强度。

Amplite 荧光法组蛋白去乙酰化酶HDAC活性检测试剂盒 绿色荧光是美国AAT Bioquest生产的用于检测HDAC的试剂盒，组蛋白去乙酰化酶（HDAC）是一类从组蛋白上的ε-N-乙酰基赖氨酸氨基酸中除去乙酰基的酶。去乙酰化恢复赖氨酸氨基酸的正电荷，这增加了组蛋白对DNA的带负电荷的磷酸骨架的亲和力。该过程通常通过阻断转录因子的进入来下调DNA转录。正在研究HDAC抑制剂作为癌症的治疗方法。

96孔板测定示例
概述
准备含有HDAC的样品（40μL）
添加HDAC抑制剂或测试化合物（10μL）
在室温或37℃孵育10-20分钟
添加HDAC Green 底物工作溶液（50μL）
在室温或37℃下孵育30-60分钟
监测Ex / Em = 490 / 525nm处的荧光强度
注意：在开始实验之前解冻所有试剂盒组分。
操作方法
1.准备工作解决方案：
1.1制备含有HDAC的测试样品：在测定缓冲液（组分B）中以1:40稀释5-10mg / mL HeLa核提取物或细胞裂解物。
注意：40μL稀释样品足以用于96孔板的一个孔。 使用前立即稀释提取物。 将溶液储存在冰上。
1.2制备HDAC抑制剂（Trichostain A）溶液的稀释液：在测定缓冲液（组分B）中以1：100稀释3mM曲古菌素A溶液（组分C），得到30μM曲古抑菌素A溶液。向每个抑制剂对照孔中加入10μL30μM曲古抑菌素A溶液。
1.3制备HDAC Green 底物工作溶液：将20μLHDACGreen 底物（组分A）和100μL信号增强剂（组分D）加入5 mL测定缓冲液（组分B）中。
注意1：稀释的HDAC Green 底物工作溶液不稳定，5 mL稀释的HDAC Green 底物工作溶液足以进行100次分析。
注意2：为每个实验准备新鲜的HDAC Green 底物工作溶液。 将重构的工作溶液保存在冰上直至使用。
2.运行HDAC分析：
2.1将40μL稀释的核提取物，酶溶液或其他HDAC样品和10μL测试化合物加入相应的微孔板孔中（参见说明书中的表1）。
对于阳性对照：用10μL测定缓冲液（组分B）加入40μL稀释的HDAC酶溶液或HeLa核提取物（来自步骤1.1）。
对于阴性对照：添加40μL稀释的HeLa核提取物（来自步骤1.1）和10μL30μM曲古抑菌素A溶液（来自步骤1.2），或使用不含HDAC活性的已知样品。
对于空白（无酶）：仅添加50μL测定缓冲液（组分B）。
2.2将板在室温或37℃下孵育10-20分钟。
注意：为了筛选HDAC抑制剂，在添加HDAC Green 底物工作溶液之前，先用HeLa核提取物或纯酶预孵育这些化合物（参见步骤2.3）
2.3将50μLHDACGreen 底物工作溶液（来自步骤1.3）加入每个孔中。 将板在室温或37℃孵育30-60分钟。
2.4 Ex / Em = 490 / 525nm时的监测荧光强度。
