样品实验方案

简要概述

1.准备黄嘌呤标准品和测试样品（50 µL）
2.添加黄嘌呤工作溶液（50 µL）
3.在室温下孵育30-60分钟
4.OD比为570/610 nm时读取吸光度增加

溶液配制

1.储存溶液配制

所有未使用的储备溶液应分为一次性使用的等分试样，并在制备后储存在-20°C下。避免重复冻融循环。
1.1 Amplite 红色底物储备液（250X）：
在Amplite 红色底物（组分A）的小瓶中加入40 µL DMSO（组分F）。注意：Amplite 红色底物在硫醇（如二硫苏糖醇（DTT）和2-巯基乙醇）的存在下不稳定。反应中DTT或2-巯基乙醇的终浓度应不高于10 µM。由于Amplite Red的pH不稳定> 8.5，因此应在pH 7 – 8（建议pH 7.4）下进行测定。

1.2 HRP储备溶液（500X）：
将100 µL测定缓冲液（组分B）加到辣根过氧化物酶（组分C）小瓶中。

1.3 黄嘌呤氧化酶（XO）储备溶液（100X）：
向小瓶黄嘌呤氧化酶标准品（组分E）中添加100 µL测定缓冲液（组分B），制成黄嘌呤氧化酶（XO）储备液（100X）。

1.4 黄嘌呤标准溶液（100 µM）：
将5 µL黄嘌呤标准品（组分D）添加到995 µL测定缓冲液（组分B）中，得到100 µM黄嘌呤标准溶液。

2.标准溶液配制

黄嘌呤标准品
用100 µM黄嘌呤标准溶液（X7）进行1：3系列稀释，以获得黄嘌呤标准液（X6-X1）。

3.工作溶液配制

将20μL的Amplite Red底物储备液（250X），10μL的HRP储备液（500X）和50μL的黄嘌呤氧化酶储备液（100X）加入5 mL的测定缓冲液中，使总体积为5.08 mL，避光。

样品操作及实验分析

表1.在透明的底部96孔微孔板（50 µL /孔）中的黄嘌呤标准品和测试样品的布局。 X =黄嘌呤标准品（X1-X7，0.1至100 µM），BL =空白对照，TS =测试样品。

表2.每个孔的试剂组成

1.根据表1和2中提供的布局，准备黄嘌呤标准品（X），空白对照（BL）和测试样品（TS）。对于384孔板，每孔使用25 µL试剂代替50 µL。

2.向黄嘌呤标准品，空白对照和测试样品的每个孔中添加50 µL工作溶液，以使黄嘌呤测定总体积为100 µL /孔。 对于384孔板，请向每个孔中添加25 µL工作溶液，总体积为50 µL /孔。

3.在避光条件下，于室温下孵育反应30至60分钟。

4.使用吸光度板读数器以570/610 nm的OD比监视信号强度。
