Pyrene is a polycyclic aromatic hydrocarbon that is well-known for its ability to intercalate into DNA. Pyrene features intense blue fluorescence. Its parameters depend strongly on the fluorophore microenvironment. Therefore, fluorescence spectra of pyrene are used for the extraction of structural information about the site surrounding pyrene. Two pyrenes in close proximity usually form excimers easily detectable by excimer fluorescence. Pyrene can also be a FRET donor to other fluorophores such as perylene.

With this phosphoramidite, pyrene can be introduced into DNA by means of automated synthesis. This pyrene phosphoramidite contains hydrocarbon moiety rigidly attached to deoxyuridine. This reagent allows attaching pyrene fragment to 5′, internal, or 3′ position (using universal support).

This amidite requires no special handling, coupling, or deprotection conditions. Recommended diluent for it is acetonitrile.

RNase B是一种高甘露糖蛋白，可作为糖苷内切酶切割N-连接糖链的阳性对照。RNase B有一个N-连接糖基化位点，可以用于SDS-PAGE凝胶电泳分析。

RNase B去糖操作步骤

1. 10 μl 反应体系中，加入2 μl 的RNase B、1μl 10X糖蛋白变性缓冲液和 5 μl H₂O。

2. 于100°C煮沸 10分钟。

3. 加入1 μl 10X GlycoBuffer 2反应缓冲液和 1 μl 10% NP-40。

4. 加入1 μl糖苷内切酶。

5. 37°C温育 1小时。

6. 将反应产物上样于10-20%的 SDS-PAGE胶。

注意事项

250 μg足够用于约20次反应。

简要概述

Annexin V-AF350标记是美国AAT Bioquest研发的用于检测细胞功能的探针，膜联蛋白是钙依赖性磷脂结合蛋白家族。它们富含真核生物，属于参与信号转导的普遍存在的细胞质蛋白家族。膜联蛋白V的优先结合配偶体是磷脂酰丝氨酸（PS），其通常保持在细胞膜的内叶（细胞质侧）。在细胞凋亡中，PS被转移到质膜的外部小叶。磷脂酰丝氨酸在细胞表面上的出现是细胞凋亡的初始/中间阶段的通用指示，并且可以在观察到形态变化之前检测到。我们的膜联蛋白V结合物是一组监测细胞功能的工具，旨在通过测量磷脂酰丝氨酸的易位来监测细胞凋亡。

分析方案

概述

用测试化合物（200μL/样品）制备细胞

添加Annexin V结合物测定溶液

室温孵育30-60分钟

用流式细胞仪或荧光显微镜分析

1.用膜联蛋白V缀合物制备和孵育细胞：

1.1制备膜联蛋白V结合测定缓冲液：10mM HEPES，140mM NaCl和2.5mM CaCl₂，pH 7.4。

1.2用测试化合物处理细胞一段所需的时间（用星形孢菌素处理的Jurkat细胞4-6小时）以诱导细胞凋亡。

1.3离心细胞，得到1-5×10⁵个细胞/管。

1.4将细胞重悬于200μL膜联蛋白V结合测定缓冲液中（来自步骤1.1）。

1.5在细胞中加入2μL膜联蛋白V结合物。

可选：在细胞内加入死细胞染色剂如碘化丙锭，用于坏死细胞。

1.6在室温下孵育30至60分钟，避光。

1.7在用流式细胞仪或荧光显微镜分析细胞之前，加入300μL膜联蛋白V结合测定缓冲液（来自步骤1.1）以增加体积（参见下面的步骤1.8）。

1.8使用流式细胞仪或荧光显微镜监测荧光强度（参见下面的步骤2或3）。

2.使用流式细胞仪分析：

通过使用具有适当过滤器的流式细胞仪来量化膜联蛋白V缀合物。

注意：粘附细胞上的膜联蛋白V结合流式细胞术分析未经常检测，因为在细胞分离或收获期间可能发生特定的膜损伤。然而，Casiola-Rosen等人先前报道了利用膜联蛋白V对贴壁细胞类型进行流式细胞术的方法。和van Engelend等人（见参考文献1和2）。

3.使用荧光显微镜分析：

3.1移取步骤1.6的细胞悬液，用膜联蛋白V结合测定缓冲液（来自步骤1.1）冲洗1-2次，然后用膜联蛋白V结合测定缓冲液（来自步骤1.1）重悬细胞。将细胞添加到盖有玻璃盖玻片的载玻片上。

注意：对于粘附细胞，建议直接在盖玻片上生长细胞。与膜联蛋白V缀合物孵育（步骤1.6）后，用膜联蛋白V结合测定缓冲液（来自步骤1.1）冲洗1-2次，并将膜联蛋白V结合测定缓冲液（来自步骤1.1）加回到盖玻片中。在玻璃载玻片上翻转盖玻片并观察细胞。在与膜联蛋白V缀合物孵育后，细胞也可以在2％甲醛中固定，并在显微镜下观察。

3.2在荧光显微镜下用适当的滤光片分析膜联蛋白V缀合物的凋亡细胞

概述

9°N™ DNA 连接酶耐热性好，能够催化磷酸二酯键的形成，使与同一互补靶 DNA 链杂交的两条相邻寡核苷酸链的 5´ -磷酸末端和 3´ -羟基末端通过磷酸二酯键相连。该连接反应只有当两条寡核苷酸链与互补靶 DNA 完全配对，并且两条寡核苷酸链之间没有空隙的条件下才可发生。9°N™DNA 连接酶以 ATP 为辅因子。该酶在 45-70℃ 范围内均有活性。

来源

纯化自重组 E. coli 菌株，其携带有从极度耐热的海底超嗜热古菌（Thermococcus sp.）9°N 菌株中克隆的连接酶基因。

R6G phosphoramidite for oligonucleotide synthesis, pure 6-isomer.

R6G (rhodamine 6G) is a xanthene dye of the rhodamine family with high fluorescence quantum yield and high molar extinction coefficient. Relative to those of fluorescein, the absorption (518 nm) and emission (542 nm) maxima of 6-R6G are shifted into the long-wave region.

R6G can be used as a FRET acceptor in DNA-sequencing and in short tandem repeat (STR) analysis. The dye is commonly used for producing TaqMan probes for quantitative PCR. In comparison with HEX and JOE dyes, R6G in combination with BHQ1 quencher has stronger quenching in molecular beacon probes, significantly reducing background fluorescence in PCR.

Usage:

Coupling: 10 minutes.

Deprotection: solution of tert-Butylamine in water (1:3 v/v) overnight at 55°С.

Aqueous ammonia and AMA (solution of 30% ammonium hydroxide/40% aqueous methylamine 1:1 v/v) must NOT be used for deprotection of the modified oligonucleotide from the solid-phase support because of complete and irreversible R6G degradation.

O-Glycosidase, also known as Endo-α-N-Acetylgalactosaminidase, catalyzes the removal of

Core 1 and Core 3 O-linked disaccharides from glycoproteins.

Neuraminidase is the common name for Acetyl-neuraminyl hydrolase ( Sialidase ). This Neura-minidase catalyzes the hydrolysis of α2-3, α2-6, α2-8 linked N-acetyl-neuraminic acid residues

from glycoproteins and oligosaccharides.

is cloned from Clostridium perfringens (2) and overexpressed in E. coli (3).

Properties and Usage

Unit Definition

One unit of O-Glycosidase is defined as the amount of enzyme required to remove 0.68 nmol of O-linked disaccharide from 5 mg of Neuraminidase digested, non-denatured fetuin in 1 hour at 37°C in a total reaction volume of 100 µl (1 unit of both O-Glycosidase and PNGase F will remove equivalent molar amounts of O-linked disaccharides and N-linked oligosaccharides, respectively).

Non-denaturing Unit Definition of O-Glycosidase:
Two fold serial dilutions of O-Glycosidase are added to a reaction mixture of 5 mg of Neura-minidase digested fetuin with 1 X GlycoBuffer 2. The reaction is then incubated at 37°C for 1 hour. O-linked disaccharide carbohydrates are determined by Morgan and Elson Assay (4). Note: Under denaturing conditions the enzyme activity is increased two-fold. This observation is substrate dependent.

Unit Definition of Neuraminidase:
One unit of Neuraminidase is defined as the amount of enzyme required to cleave > 95% of the terminal α-Neu5Ac from 1 nmol Neu5Acα2-3Galβ1-3GlcNAcβ1-3Galβ1-4Glc-7-amino-4-methyl- coumarin (AMC), in 5 minutes at 37°C in a total reaction volume of 10 µl.

Notes

1.Since O-Glycosidase is inhibited by SDS, it is essential to have NP-40 in the reaction mixture. It is not known why this non-ionic detergent counteracts the SDS inhibition at the present time. Double digest with Endo H must have NP-40 present (NP-40 does not inhibit Endo H).

2.To deglycosylate a native glycoprotein, longer incubation time as well as more enzyme may be required.
