产品介绍
碘化丙啶（PI）属于与溴化乙锭相同的化学类别。与溴化乙锭的情况一样，在与核酸结合后的荧光增强了20-30倍。荧光激发最大值红移为30-40nm，荧光发射最大值蓝移为15nm左右。PI通常用于鉴定细胞群中的死细胞，并用作多色荧光技术中的复染剂。它还可用于区分坏死细胞，凋亡细胞和正常细胞。它适用于荧光显微镜，流式细胞仪和荧光仪。

染色细胞示例
PI染色通常在其他染色后进行，它只能染死细胞，以下程序适用于大多数细胞类型。生长培养基，细胞密度，细胞类型和其他因素可能会影响染色影响染色情况。玻璃器皿上的残留的洗涤剂也可能影响许多生物染色，导致在没有细胞存在的溶液中出现明亮的染色物质。
1）制备1-10mM DMSO储备溶液。如果储存在-20°C，DMSO储备液可以保存6个月。
2）选择适合您样品的固定方案。
3）通过离心沉淀细胞，并将细胞重悬于缓冲盐溶液或培养基中，在pH 7.4下进行最佳染料染色。培养物中的贴壁细胞可以在盖玻片上或细胞培养孔中原位染色。
4）添加PI进行染色（浓度为0.5-5μM之间），并将其孵育15至60分钟。
注意：在最初的实验中，最好在整个建议的范围内尝试几种染料浓度，以确定产生最佳染色的浓度。
