Pluronic F-127是一种非离子表面活性剂,100%活性,对低浓度细胞无毒,常用于染料AM酯,如Indo-1 AM,Fura-2 AM,Calcein AM,Fluo-3 AM,Fluo-4 AM,Quest Fluo-8 AM和Quest Rhod-4 AM等,以改善其水溶性。 Pluronic F-127也可用于分散其他亲脂性探针。应进行适当的控制以确保Pluronic F-127不会改变细胞的膜特性。为方便起见,我们还提供20%Pluronic F-127 DMSO溶液(Cat#20052)和10%Pluronic F-127水溶液(Cat#20053)。

操作说明
注意:以下是我们推荐的方案。该方案仅提供指南,实际应根据您的具体需求进行修改。
1.将1克Pluronic F-127溶于10毫升蒸馏水中,制成10%(w / v)原液,或2克Pluronic F-127在10毫升无水二甲基亚砜(DMSO)中制成20%(w / v)储备溶液。这些可能需要在40至50℃的温度下加热约30分钟。在室温下储存溶液。
注意:不要冷却或冷冻Pluronic F-127溶液,因为它可能会沉淀。如果观察到沉淀,可以通过加热至37℃使沉淀物溶解并在使用前涡旋。
2.将10%或20%Pluronic®F-127储备液(来自步骤1)稀释到细胞上样缓冲液中,如Hanks和20 mM Hepes缓冲液(HHBS),稀释度为1:1000至1:500,以达到0.02至0.04%的工作溶液。
3.然后将AM酯的DMSO储备溶液稀释到0.02至0.04%的工作溶液(来自步骤2)中,以获得1μM至10μM的最终AM酯浓度。
注意:Pluronic F-127的最终浓度通常保持在0.08%或以下。
4.将细胞在所需温度下孵育10分钟至1小时。
注意:通常,希望使用所需的最少量的AM酯来获得足够的荧光信号噪声水平。
5.标记后,在开始实验之前用HHBS或新鲜培养基洗涤细胞。
