几乎所有质膜在其上都具有电势,内部通常相对于外部为负。信号是通过打开或关闭膜上某一点的离子通道而产生的,从而在膜电位上产生局部变化。电场的这种变化会很快受到膜中相邻或较远的离子通道的影响。然后,由于电势变化,这些离子通道可以打开或关闭,从而产生信号。 Cell Meter 荧光细胞电位检测试剂盒使用FRET对(VSB 405和VSR 555)来检测细胞膜电位的变化。亲脂性VSB 405主要位于脂质膜的外层,而VSR 555的定位对细胞膜电位敏感。在静止状态下,的内层具有相对负的电势,从而使VSR 555主要位于细胞膜的外层附近,因此靠近蓝色荧光VSB405,从而导致从蓝色(VSB 405)到蓝色荧光的有效转移。红色(VSR 555)。当细胞去极化时,VSR 555易位到细胞膜的内层,从而分离FRET对并破坏FRET。蓝色/红色荧光比与细胞电位成正比。该检测法可用于筛选调节离子通道的化合物。
