新型钙离子荧光探针Calbryte-520L AM是美国AAT Bioquest生产的钙离子荧光探针,细胞内钙通量测定法是监测信号转导途径和G蛋白偶联受体(GPCR)和钙通道靶标的高通量筛选的一种广泛使用的方法。随后在1989年推出Fluo-3,随后开发了具有改善的信噪比的Fluo-4,Fluo-8和Cal-520,并成为共聚焦显微镜,流式细胞术和高通量筛选应用中广泛使用的Ca2 +指示剂。但是,Fluo-4仍然存在一些严重问题。例如,对于Fluo-3,在所有大多数使用Fluo-4 AM进行的细胞内钙测定中,都需要丙磺舒以防止细胞加载的Fluo-4泄漏出细胞。丙磺舒与基于Fluo-4的钙测定法的结合使用会损害测定结果,因为众所周知丙磺舒具有多种复杂的细胞作用。 Calbryte 520L,AM是一种新型的荧光且可渗透细胞的钙指示剂。像其他染料AM细胞加载一样,Calbryte 520L AM酯不发荧光,一旦进入细胞内部,它就会被细胞内酯酶水解并被激活。激活的指示剂是一种极性分子,不再能够自由扩散通过细胞膜,基本上被困在细胞内部。 Calbryte 520L对钙离子的亲和力低,Kd约为100 uM。 Calbryte 520L在高浓度钙离子存在下产生明亮的荧光信号。它具有与Fluo-4相同的激发和发射波长,因此相同的Fluo-4测定设置可轻松应用于基于Calbryte 520L的钙测定。金畔生物是AAT Bioquest的中国代理商,为您提供最优质的新型钙离子荧光探针Calbryte-520L AM。
