吖啶橙是一种核酸选择性荧光阳离子染料,可用于细胞周期测定。 它是细胞可渗透的,并通过插入或静电吸引力与DNA和RNA相互作用。当与DNA结合时,它与荧光素在光谱上非常相似,激发最大值为502nm,发射最大值为525nm(绿色)。当它与RNA结合时,激发最大值移至460nm(蓝色),发射最大值移至650nm(红色)。染料通常用于落射荧光显微镜。

染色细胞示例
以下程序适用于大多数细胞类型。生长培养基,细胞密度,其他细胞类型的存在和其他因素可能影响染色。玻璃器皿上的残留洗涤剂也可能影响许多生物的真实或明显染色,导致在有或没有细胞存在的溶液中出现明亮染色的物质。
1)制备1-10mM DMSO储备溶液。如果储存在-20°C,DMSO储备液可以保存6个月。
2)使用适合您样品的固定方案。
3)通过离心沉淀细胞,并将细胞重悬于缓冲盐溶液或培养基中,在pH 7.4下进行最佳染料染色。培养物中的贴壁细胞可以在盖玻片上或细胞培养孔中原位染色。
4)使用1和10μM之间的浓度添加AO染色并孵育15至60分钟作为指导。
注意:在最初的实验中,最好在整个建议的范围内尝试几种染料浓度,以确定产生最佳染色的浓度。
